An ELISA (Enzyme-Linked Immunosorbent Assay) is a versatile analytical technique used to measure the concentration of proteins or other biomolecules. The method relies on the specific interaction between an antibody and its corresponding antigen. Detection is achieved through an enzyme-catalyzed reaction, producing a measurable signal—often quantified with a spectrophotometer. By using calibrators or standards, ELISA enables accurate quantitative measurements. Its accessibility, reliability, and broad applicability to samples such as serum and plasma make ELISA a widely used tool in both research and clinical diagnostics.

Common ELISA formats include direct, indirect, competitive, and sandwich assays:

  • Direct ELISA: The antigen is immobilized on a plate, and a single enzyme-labeled primary antibody binds directly to the target.
  • Indirect ELISA: Detection involves an unlabeled primary antibody followed by an enzyme-conjugated secondary antibody.
  • Competitive ELISA: Sample antigen and a labeled antigen compete for a limited number of antibody-binding sites. Higher concentrations of sample antigen reduce the signal from the labeled antigen.
  • Sandwich ELISA: The target antigen is captured by an immobilized antibody, followed by binding of a second enzyme-labeled antibody to a different epitope. This format is particularly sensitive and specific, making it ideal for detecting low-abundance biomarkers in complex biological samples.

The enzyme-substrate reaction generates a color signal proportional to antigen concentration, calibrated using standards or samples with known values.
