Äkta Pure 25 System

The AKTA Pure 25 is an advanced chromatography platform widely used for the separation and purification of proteins, peptides, nucleic acids, and other biomolecules. Designed for both laboratory and small‑scale production, this system supports a broad range of chromatographic techniques, including ion‑exchange, size‑exclusion, affinity, and other methods, enabling precise purification based on molecular properties such as size, charge, and interaction characteristics.

The ÄKTA Pure 25 system features a modular, flexible architecture that can be adapted to your research needs. It offers a high flow rate range (typically 0.001–25 mL/min) and can reach pressures up to 20 MPa, making it suitable for both analytical and preparative tasks. With real‑time monitoring of UV absorbance, conductivity, and pH, the system provides detailed insights into each purification run.

At the front of the instrument is the “wet” working area, where fluidic connections for buffers and columns can be adjusted as needed. All components in this section can be rinsed with water, and small chromatography columns can be easily mounted using clamp holders. A drip tray located underneath captures any spills or leaks. Larger columns are attached on the left side of the system, while the right side houses the electrical components.

The pump section consists of two precision pumps that can be operated individually or in combination to form gradients, allowing seamless transition between buffers during separation. Typically, one pump handles multiple inlet lines for wash or binding buffers, while the other is primarily used for elution buffers, both controlled through the intuitive UNICORN software, which enables method creation, run monitoring, and data analysis.
