At Biocult, the AcuSyst-Jr and Maximizer bioreactor systems are equipped with fully integrated culture sets. The Maximizer system offers the most versatile setup, making it ideal for a wide range of cell culture applications.

A typical two-artificial-kidney culture set consists of two separate circuits:

    • Fresh medium is pumped into the IC mixing chamber and circulated through the GEX system.

    • The medium is aerated and flows through the artificial kidneys, with a portion removed via the outflow pump.

    • This circulation allows precise control of the medium composition based on glucose and pH, ensuring optimal culture conditions.

2. Expansion Circuit (EC – cell-containing, yellow)

    • This circuit contains the cells, which are circulated through the artificial kidneys via an external pump.

    • Supplements (medium with serum) can be directly added without affecting the IC.

    • The culture suspension (‘harvest’) is collected in a sterile bottle for downstream use.

    • Any pellet formation can be cleared using the dedicated “cell removal” line to prevent blockage.

Hollow Fiber System (“Artificial Kidney”)

In the artificial kidney, fresh, pH-adjusted medium containing nutrients enters one side, while medium carrying waste exits the other. This process, driven by the Starling effect, arises from the pressure difference across the fibers and the flow along the length of the kidney. It efficiently supplies the cells between the fibers with nutrients and oxygen while simultaneously removing metabolic waste.

To maintain a homogeneous environment and optimal conditions for the cells, cell cycling is used. This technique alternates backpressure between the EC and IC mixing chambers over a defined time and fluid-level difference, ensuring even nutrient delivery, effective waste removal, and uniform distribution throughout the artificial kidney. This process also allows cultivation at higher cell densities.

Process Monitoring and Control

  • Temperature: Monitored and controlled via an integrated temperature probe.
  • pH: Measured and regulated using a pH probe in the circulating medium.
  • Dissolved Oxygen (DO): Monitored and controlled with a DO probe.
  • Glucose and pH: External measurements are performed by sampling conditioned medium from the IC under sterile conditions.
